(Biolegend-302212)Human peripheral blood lymphocytes were stained with CD19 (clone 4G7) (filled histogram) APC or mouse IgG1, κ isotype control (open histogram) APC.
The antibody was purified by affinity chromatography and conjugated with APC under optimal conditions.
Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is 5 µl per million cells in 100 µl staining volume or 5 µl per 100 µl of whole blood.
CD19 is a 95 kD type I transmembrane glycoprotein also known as B4. It is a member of the immunoglobulin superfamily expressed on B-cells (from pro-B to blastoid B cells, absent on plasma cells) and follicular dendritic cells. CD19 is involved in B cell development, activation, and differentiation. CD19 forms a complex with CD21 (CR2) and CD81 (TAPA-1), and functions as a BCR co-receptor.
APC anti-human CD19是一類以anti-human CD19為抗原的APC多克隆抗體,可以特異性結合human CD19,主要用于檢測CD19的Western Blot、IHC-P、IF、ELISA、Co-IP等多種免疫學實驗。
CD19檢測以免疫組織化學方法為主,利用抗原抗體發(fā)生特異性結合的特點,通過特異性標記抗體與CD19抗原結合,進行孵育并通過染色鏡檢、儀器計數(shù)等方式對標記進行識別,從而達到定性或定量檢測的目的。流式細胞術可檢測全體B細胞及毛細胞白血病細胞上的CD19,免疫組織染色法可檢測全體B細胞及濾泡樹狀細胞上的CD19。其他方法有免疫熒光染色、免疫金銀染色等。
CD19是分子量為95kDa的糖蛋白,表達于前B細胞和成熟的B細胞膜表面,與B細胞的活化調(diào)節(jié)和發(fā)育調(diào)節(jié)相關,在T細胞和正常的粒細胞上無表達。此抗體主要標記正常B細胞和腫瘤性B細胞。
CD19是參與B細胞活化與增殖的重要膜抗原之一,是所有B細胞共有的表面標志,B細胞活化后不消失,是最重要的B細胞標記因子,同時CD19也是B細胞表面的傳達信號復合體的構成部分,CD19的細胞外部分同其他膜抗原結合進行信號傳導。
APC anti-human CD19用于CD19~+CD24~(hi)CD38~(hi)Breg在PBC患者外周血中表達及對Th1細胞分化影響的研究。
