向1000 mL干燥的三口燒瓶中依次加入60 g(0.416 mol)異丁酰乙酸甲酯、120 g(1.288 mol)苯胺和6 g 4-二甲基氨基吡啶(DMAP)。開啟攪拌,緩慢升溫至內(nèi)溫100℃,保持此溫度反應(yīng)16小時(shí)。反應(yīng)過程中,通過分餾裝置收集生成的甲醇。反應(yīng)結(jié)束后,將體系降溫至70℃,減壓蒸餾回收未反應(yīng)的苯胺,共回收74.7 g苯胺,回收率為92%。剩余反應(yīng)液濃縮至約100 g后,加入1 kg水,冷卻至30℃并攪拌結(jié)晶5小時(shí)以上。過濾,用水洗滌濾餅3次,干燥后得到白色固體產(chǎn)物82.3 g。經(jīng)HPLC分析,產(chǎn)物純度為99.8%,收率為96.4%。
