在500 mL三頸圓底燒瓶中,將Ni(NO3)2·6H2O(12.6 g,43.33 mmol)溶解于甲醇(200 mL)中。向該溶液中加入5-溴-8-硝基-N-甲基異喹啉碘化物(33.33 g,84.38 mmol)。隨后,分批加入NaCNBH3(10.6 g,168.68 mmol)。將反應混合物在室溫下攪拌5小時,反應進程通過薄層色譜(TLC)監(jiān)測(展開劑比例為EtOAc:PE = 1:5)。反應完成后,使用旋轉(zhuǎn)蒸發(fā)器在減壓下濃縮反應液。將殘余物溶解于800 mL水中,并用5% NaOH溶液調(diào)節(jié)pH至8-10。過濾后,用800 mL EtOAc萃取水相兩次,合并有機層并用Na2SO4干燥。最后,通過柱色譜法(洗脫劑比例為EtOAc:PE = 1:5)純化殘余物,得到19.3 g(產(chǎn)率83%)的5-溴-2-甲基-8-硝基-1,2,3,4-四氫異喹啉,為黃色固體。
