在氮?dú)獗Wo(hù)下,將5-甲氧基苯并[b]噻吩(實(shí)施例20a,1.5g,9.14mmol,1.0當(dāng)量)溶于二氯甲烷(20mL)中,冷卻至-10℃。緩慢滴加三溴化硼(1M in DCM,36.6mL,36.6mmol,4.0當(dāng)量)。保持該溫度攪拌反應(yīng)30分鐘后,緩慢升溫至室溫繼續(xù)反應(yīng)1小時(shí)。反應(yīng)完成后,在-5℃下用甲醇淬滅反應(yīng)。用二氯甲烷(50mL)和水進(jìn)行萃取,有機(jī)層用飽和亞硫酸氫鈉水溶液洗滌,無水硫酸鈉干燥后濃縮。殘余物通過快速柱色譜(石油醚:乙酸乙酯=4:1)純化,得到目標(biāo)產(chǎn)物苯并[b]噻吩-5-醇(實(shí)施例20b,1g,產(chǎn)率73%,黃色固體)。質(zhì)譜分析顯示[M + 1] + = 151.0。
