步驟A-合成7-甲基-5,7-二氫-6H-二苯并[b,d]氮雜卓-6-酮:在干燥的圓底燒瓶中加入氫化鈉(0.295 g,7.46 mmol)和9.0 mL無水DMF,攪拌至完全溶解。隨后加入5,7-二氫-6H-二苯并[b,d]氮雜卓-6-酮(1.3 g,6.22 mmol),反應(yīng)混合物在60℃下攪拌1小時(shí)。然后,緩慢滴加碘甲烷(1.16 mL,18.6 mmol),避光條件下繼續(xù)攪拌17小時(shí)。反應(yīng)完成后,將混合物冷卻至室溫,用二氯甲烷和水的混合液稀釋,依次用NaHSO4溶液和水洗滌,有機(jī)相用無水Na2SO4干燥。減壓濃縮后,通過快速柱色譜(硅膠,氯仿作為洗脫劑)純化,得到0.885 g(產(chǎn)率63%)目標(biāo)化合物5-甲基-5H-二苯并[b,d]氮雜卓-6(7H)-酮,為無色固體。核磁共振氫譜(1H-NMR, CDCl3)數(shù)據(jù):δ= 7.62(d, 2H),7.26-7.47(m, 6H),3.51(m, 2H),3.32(s, 3H)。元素分析:C15H13NO(分子量=223.27);質(zhì)譜(MH+)223。計(jì)算值:C, 80.69;H, 5.87;N, 6.27。實(shí)測(cè)值:C, 80.11;H, 5.95;N, 6.23。
