以4-(3-吡啶基)-3-丁炔-1-醇為原料合成4-(3-吡啶基)-1-丁醇的一般步驟:將4-(3-吡啶基)-3-丁炔-1-醇(0.32 g,2.2 mmol)溶于EtOH(44 mL)中,通過裝有10% Pd/C催化劑的H-Cube?流動氫化反應(yīng)器進(jìn)行氫化反應(yīng)(流速:1.0 mL/min;壓力:1.0 bar;溫度:3.5°C)。反應(yīng)完成后,將輸出溶液濃縮至干,得到油狀殘余物(0.31 g)。通過常規(guī)硅膠快速色譜法(洗脫劑:CH2Cl2/MeOH,95:5)純化,得到純的4-(3-吡啶基)-1-丁醇(0.294 g,收率89%),為無色液體。Rf = 1.16 mm。MS(ESI)m/z:152 [M-H]-。1H NMR(DMSO-d6)δ:8.43(m,1H),8.38(dd,1H,J = 4.8, 1.5 Hz),7.61(dt,1H,J = 7.8, 2.1 Hz),7.30(dd,1H,J = 7.8, 4.8 Hz),4.37(t,1H,J = 5.2 Hz),3.41(q,2H,J = 5.2 Hz),2.59(t,2H,J = 7.7 Hz),1.64-1.55(m,2H),1.47-1.38(m,2H)。
