以[4-(4,6-二氯嘧啶-2-基磺酰基)苯基]甲胺環(huán)丙羧酸(3.0g,8.85mmol)和3-氨基-5-甲基-1H-吡唑(0.94g,9.7mmol)為原料,在DMF(16mL)中混合,加入二異丙基乙胺(1.25g,9.7mmol)和碘化鉀(1.76g,10.6mmol)。將反應(yīng)混合物在50℃下攪拌6小時。反應(yīng)完成后,減壓蒸發(fā)去除溶劑,殘余物通過乙酸乙酯重結(jié)晶純化。得到目標(biāo)產(chǎn)物N-[4-[[4-氯-6-(5-甲基-2H-吡唑-3-氨基)嘧啶-2-基]磺?;鵠苯基]氨基環(huán)丙羧酸(2.0g,收率91%),為白色固體。LC-MS(m/z):401 [M + 1]+;1H NMR(DMSO-d6)δ:0.82(d,J = 6.6 Hz,4H),1.85(m,1H),1.99(s,1H),5.24(bs,1H),6.47(bs,1H),7.55(m,2H),7.71(m,2H),10.22(s,1H),11.95(s,1H)。
