| In vitro | CLK8 showed no toxicity to U2OS cells with a maximum concentration of 40 μM, maintaining cell viability above 80%. Similarly, CLK8 enhanced the amplitude of Bmal1-d Luc signal in U2OS and NIH 3T3 cells in a dose-dependent manner, and no circadian oscillations were observed in both cell lines[1]. |
| In vivo | CLK8 was administered intraperitoneally to mice at a concentration of 25 mg/kg. The results indicated that CLK8 decreased the levels of CLOCK in the whole-cell lysate of mouse liver, while the levels of BMAL1 and CRY1 remained unchanged [1]. |
