| Synthesis | Step 2. Synthesis of furo[2,3-d]pyrimidin-4-amine (C2). 2-Aminofuran-3-carbonitrile (C1, 100 mg, 0.925 mmol) was dissolved in formamide (2 mL) and the reaction mixture was heated at 120 °C overnight. After completion of the reaction, the mixture was cooled to room temperature and subsequently partitioned between water and ethyl acetate. The aqueous layer was extracted with ethyl acetate and dichloromethane, respectively. The organic layers were combined, dried with anhydrous magnesium sulfate, filtered and concentrated in vacuum to give a yellow solid product.
