來源于S-腺苷蛋氨酸(S-adenosylmethioine,AdoMet)的甲基在特異的轉(zhuǎn)甲基酶作用下可轉(zhuǎn)移到各種受體分子(如:蛋白、核酸和小分子)上,S-腺苷高半胱氨酸(S-adenosylhomocysteine,AdoHcy)是所有AdoMet依賴的轉(zhuǎn)甲基反應(yīng)的產(chǎn)物,而S-腺苷高半胱氨酸水解酶(EC3.3.1.1)對AdoMet依賴的轉(zhuǎn)甲基反應(yīng)起著重要的調(diào)節(jié)作用。目前所知,在真核生物中S-腺苷高半胱氨酸水解酶催化S-腺苷高半胱氨酸水解生成腺苷(adenosine,Ado)和高半胱氨酸(homocysteine,Hcy)是S-腺苷高半胱氨酸代謝的唯一途徑。S-腺苷高半胱氨酸作為AdoMet依賴的轉(zhuǎn)甲基反應(yīng)的競爭性抑制劑,當(dāng)S-腺苷高半胱氨酸水解酶受到抑制時(shí),將導(dǎo)致細(xì)胞內(nèi)S-腺苷高半胱氨酸的積累,引起S-腺苷高半胱氨酸與AdoMet濃度比升高,進(jìn)而抑制AdoMet依賴的轉(zhuǎn)甲基反應(yīng),最終由于細(xì)胞內(nèi)S-腺苷高半胱氨酸水平失控而產(chǎn)生細(xì)胞毒性。
