Immunofluorescent analysis of 4% paraformaldehyde-fixed, 0.1% Triton X-100 permeabilized HeLa (Human epithelial cells from cervix adenocarcinoma) cells labeling JNK1+JNK2+JNK3 with ab179461 at 1/250 dilution, followed by Goat anti-rabbit IgG (Alexa Fluor® 488) (ab150077) secondary antibody at 1/400 dilution (green). Confocal image showing both cytoplasmic and nuclear staining on HeLa cells. The nuclear counterstain is DAPI (blue). Tubulin is detected with ab7291 (anti-Tubulin mouse mAb) at 1/500 dilution and ab150120 (AlexaFluor®594 Goat anti-Mouse secondary) at 1/500 dilution (red).
The negative controls are as follows:- -ve control 1: - ab179461 at 1/250 dilution followed by ab150120 (AlexaFluor®594 Goat anti-Mouse secondary) at 1/500 dilution. -ve control 2: - ab7291 (anti-Tubulin mouse mAb) at 1/500 dilution followed by ab150077 (Alexa Fluor®488 Goat Anti-Rabbit IgG H&L) at 1/400 dilution.
C-Jun N末端激酶(JNK)也稱為絲裂原激活的蛋白激酶(MAPK8)。它屬于應(yīng)激激活蛋白激酶的MAPK超家族。MAPK是絲氨酸-蘇氨酸蛋白激酶,可響應(yīng)多種細(xì)胞外刺激而被激活,并介導(dǎo)從細(xì)胞表面到細(xì)胞核的信號轉(zhuǎn)導(dǎo)。JNK參與多種細(xì)胞過程,例如增殖,分化,轉(zhuǎn)錄調(diào)節(jié)和發(fā)育。JNK途徑受壓力和炎癥信號的刺激。由于差異的mRNA剪接,JNK被表達(dá)為十種不同的亞型。主要形式為JNK1和JNK2。 JNK123能夠使c-Jun(一種轉(zhuǎn)錄調(diào)節(jié)因子,屬亮氨酸拉鏈家族成員)氨基末端活性區(qū)Ser63和Ser73發(fā)生磷酸化的蛋白激酶。是促分裂原活化的蛋白激酶超家族中的一個家族,包括JNK1、JNK2、JNK3,當(dāng)其活化模體Thr-Pro-Tyr內(nèi)的Thr、Tyr被JNKK1、JNKK2磷酸化后而激活,活化酶可磷酸化c-Jun等多種蛋白質(zhì),從而影響細(xì)胞凋亡、炎癥和腫瘤的發(fā)生。JNK123抗體是一類可以特異性結(jié)合JNK123的多克隆抗體,主要用于檢測JNK123的Western Blot、IHC-P、IF、ELISA、Co-IP等多種免疫學(xué)實(shí)驗(yàn)。雙抗體夾心法測定標(biāo)本中JNK123水平。用純化的JNK123抗體包被微孔板,制成固相抗體,往包被單抗的微孔中依次加入JNK123,再與HRP標(biāo)記的JNK123抗體結(jié)合,形成抗體-抗原-酶標(biāo)抗體復(fù)合物,經(jīng)過徹底洗滌后加底物TMB顯色。TMB在HRP酶的催化下轉(zhuǎn)化成藍(lán)色,并在酸的作用下轉(zhuǎn)化成最終的黃色。顏色的深淺和樣品中的JNK123呈正相關(guān)。用酶標(biāo)儀在450nm波長下測定吸光度(OD值),通過標(biāo)準(zhǔn)曲線計算樣品中JNK123濃度。
