以3-氨基-5-甲基-1H-吡唑和2,4,5-三氯嘧啶為原料合成2,5-二氯-N-(5-甲基-1H-吡唑-3-基)-4-氨基嘧啶的一般步驟如下:向5-甲基-1H-吡唑-3-胺(4.00 g,41.2 mmol)的無(wú)水乙醇(100 mL)溶液中加入三乙胺(12.51 g,123.6 mmol)和2,4,5-三氯嘧啶(7.56 g,41.2 mmol)。將反應(yīng)混合物在室溫下攪拌16小時(shí),隨后在真空下濃縮。通過(guò)硅膠柱色譜法(石油醚/乙酸乙酯,v/v = 1/1)純化殘余物,得到目標(biāo)化合物2,5-二氯-N-(5-甲基-1H-吡唑-3-基)-4-氨基嘧啶,為淺黃色固體(9.00 g,收率89.5%)。質(zhì)譜(ESI,正離子模式)m/z:244.1 [M + H]+;核磁共振氫譜(600 MHz,DMSO-d6)δ(ppm):12.32(寬單峰,1H),9.70(單峰,1H),8.33(單峰,1H),6.28(單峰,1H),2.25(單峰,3H)。
