Uses4-(N-BOC-N-methylamino)cyclohexanol can be used as a pharmaceutical synthesis intermediate.
Application4-(N-BOC-N-methylamino)cyclohexanol (30 g, 149.1 mmol, 1 eq.), triethylamine (22.87 mL, 164 mmol, 1.1 eq.), and N,N-dimethylpyridine-4-amine (DMAP) (1.83 g, 14.98 mmol, 0.1 eq.) were dissolved in anhydrous dichloromethane (500 mL) and cooled to 0 °C in an ice bath. Methanesulfonyl chloride (12.12 mL, 156.6 mmol, 1.05 equivalents) was added dropwise. After the addition was complete, the reaction mixture was warmed to room temperature and stirred for 16 hours. The reaction mixture was washed with water (3 × 100 mL) and saturated sodium bicarbonate (3 × 100 mL). The combined washings were back-extracted with dichloromethane. The combined organic compounds were dried over Na2SO4 and concentrated to give 40.83 g (146.2 mmol) of 1-(tert-butyloxycarbonyl)piperidine-4-ylmethanesulfonate (compound 1013, 98% yield), which was a grayish-white solid and could be used without further processing.
