| Uses | 2,4-Dichlorofuro[3,2-d]pyrimidine is a furopyrimidine derivative used in the preparation of phosphoinositide 3-kinase inhibitor. | | Synthesis | Furo[3,2-d]pyrimidine-2,4-diol (7J) (1.52 g, 10 mmol) was added to N,N-dimethylaniline (1 mL, 8 mmol) and trichlorophosphorus (0.9 mL, 9.65 mmol) as a starting material and the reaction was carried out for 3 hours at 130°C. The reaction was carried out at 130°C for 3 hours. Upon completion of the reaction, the mixture was cooled to room temperature and carefully quenched with ice water. The resulting solid product was collected by filtration, washed with water and dried under vacuum to afford 2,4-dichlorofuro[3,2-d]pyrimidine (7K) (1.02 g, 54% yield) as a light brown solid. The melting point of the product was 107.3°C. The NMR hydrogen spectrum (300 MHz, CDCl3) data were as follows: δ 8.08 (d, J = 2.2 Hz, 1H), 7.02 (d, J = 2.2 Hz, 1H). | | References | [1] Patent: WO2011/79230, 2011, A2.
