17-DMAG is a potent HSP90 inhibitor with an IC50 of 62 nM in cell-free trials. 17-DMAG treated at 5mg/kg or 25mg/kg three times per week significantly reduced TMK-1 graft tumor growth by significantly reducing vascular area and proliferating tumor cell count. Consistent with inhibiting FAK signaling, 17-DMAG treated mice at a dose of 25mg/kg three times per week significantly inhibited tumor growth and metastasis of ME180 and SiHa grafted tumors. 17-DMAG treated TCL1-SCID transplanted mouse model at a dose of 10mg/kg for 16 days, significantly reduced the number of white blood cells and extended the life span.
