| Description | AZ 5704 (compound 74) is a selective and orally active ATM inhibitor (IC50 = 0.6 nM), which can enhance the anti-tumor activity of irinotecan in the SW620 xenograft model. |
| In vivo | Method: In immunodeficient SW620 (colorectal cell line) tumor-bearing mice, administer Irinotecan (50 mg/kg, intraperitoneal injection, once daily on Day 1) and AZ 5704 (50 mg/kg, oral, twice daily on Days 2 and 4; no dosing on Days 5 and 7).
Results: AZ 5704 monotherapy showed no significant efficacy. However, after 3 weeks of combination therapy with Irinotecan, tumor growth in the SW620 xenograft model was markedly reduced, with greater efficacy than Irinotecan monotherapy. |
