| In vitro | A series of novel high-affinity, reversible inhibitors of NLRP3 activation were discovered by screening a DNA-encoded library, and a potent lead compound, BAL-0028, was identified directly from the screen with an IC50 value of 25 nM; SPR studies showed that BAL-0028 binds tightly to the NACHT domain of NLRP3 (KD range of 104-123 nM); CADD analysis of the interaction of BAL-0028 with the NLRP3 NACHT domain showed that the binding site is distinct from that of ADP and MCC950 and includes site-specific interactions.
