| In vitro | Methods: Synthetic O-GlcNAc peptides (containing Ser/Thr/Tyr modifications) were labeled with GalT1(Y289L) enzyme, followed by copper-catalyzed click reaction with DADPS-biotin-propargylamide (10 mM) for 2 hours; after enrichment, samples were cleaved with 10% FA for 1 hour and analyzed by nanoUPLC-MS/MS. Results: The DADPS probe successfully enriched all types of O-GlcNAc peptides with higher intensity than the Dde probe, suitable for O-GlcNAc proteomic analysis of complex samples. [1] |
