| Description | Evans blue (C.I. 23860) is an effective inhibitor of glutamate uptake by synaptic vesicles and also acts as an antagonist of AMPA/kainate receptors. Evans blue is a water-soluble azo fluorescent dye that binds to serum albumin and is used to detect vascular permeability, blood-brain barrier integrity, and permeability. |
| In vitro | Methods: Human glioblastoma U87MG cells (FAP-positive) were treated with 1??Lu-Evans blue-FAPI-B1, B2, B3, B4 (37 kBq) tracers added to each well. Blocking group additionally received 10 μg/well FAPI-02. Treated for 0.5, 1, 2, 4, 8, 24 h. Radioactivity measured by γ-counter to calculate cellular uptake rate (%). Results: All tracers showed time-dependent uptake peaking at 24 h. Blocking agent reduced uptake rate, confirming binding specificity. [1] Methods: HT22 mouse hippocampal neuronal cells were preincubated with Evans blue (1 mM) for 2 hours, then stimulated with glutamate (100 μM) for 30 minutes. ATP concentration in cell culture supernatant was measured by bioluminescence assay. Results: Glutamate stimulation increased ATP release in HT22 cells, and Evans blue pretreatment completely blocked this effect. [2] |
| In vivo | Methods: U87MG tumor-bearing nude mice (initial tumor size ~100 mm3) received a single tail vein injection of different doses of the radiotracer 1??Lu-Evans blue-FAPI-B1 (7.4, 18.5, 30 MBq/mouse). Tumors were observed for 22 days post-treatment. Results: All treatment groups showed significant inhibition of tumor growth compared to the saline control group. [1] Methods: Male SD rats underwent L5 spinal nerve ligation (SNL) model. Intrathecal Evans blue (50 μg/rat) was administered as a single dose starting 7 days post-surgery, with observations conducted for 7 days. Results: Evans blue increased the mechanical pain threshold in a dose-dependent manner, with 50 μg showing optimal efficacy. Analgesic effects persisted for approximately 5 days.[2] |
