| Description | Methyl arachidonate functions as a protein kinase C activator and also serves as an orally active substrate that can be hydrolyzed via esterase-mediated mechanisms. Its mechanism of action is indirectly mediated by eicosanoid metabolites generated through the arachidonic acid metabolic pathway. At lower concentrations, the effects of methyl arachidonate are primarily dependent on cyclooxygenase-derived products, whereas at higher concentrations, metabolites from the lipoxygenase pathway predominate. Methyl arachidonate has applications in research related to lipodystrophy. |
| In vitro | Method: Intact human platelets were exposed to methyl arachidonate, including 50 μM treatment, and protein kinase C translocation or activation was measured over time. Purified brain protein kinase C was also tested in a cell-free assay. Result: Methyl arachidonate activated protein kinase C in intact platelets with activity comparable to arachidonic acid and reached a plateau at approximately 2 min at 50 μM. It had little direct activity on purified protein kinase C, indicating that platelet activation depended mainly on metabolic conversion products.[1]. Method: Methyl arachidonate and structurally related fatty acids were applied to purified protein kinase C and intact platelets. Protein kinase C stimulation, platelet aggregation and secretion were assessed. Result: In this experimental system, methyl arachidonate did not directly stimulate protein kinase C and did not induce platelet aggregation or secretion, providing a negative result distinct from that observed with free unsaturated fatty acids.[2]. |
