MRTX1133 is a powerful, selective and non -covalent KRAS G12D inhibitor, showing that the KD KD of KRAS G12D is 0.2 PM. MRTX1133 can prevent the formation of SOS1 catalytic nucleotide exchange and/or KRAS G12D/GTP/RAF1 complexes, thereby inhibiting the mutant KRAS dependent signal transfer. MRTX1133 KRAS G12D mutant tumor cells have selective inhibitory and do not suppress KRAS wild cells. MRTX1133 has Nonol -level activity in cell experiments, and shows significant in vivo effects in the tumor model containing KRAS G12D mutations.
