Streptavidin -HRP
Streptavidin is a protein isolated from Streptomyces avidinii, possessing biotin-binding characteristics similar to avidin. The molecular weight of Streptavidin is c.a. 60 kDa, comprising four subunits, each capable of binding one molecule of biotin. Biotin is a water-soluble vitamin with a molecular weight of 244 Da. Streptavidin exhibits extremely high binding affinity to biotin (Kd 10-15 ).
Streptavidin-HRP conjugate combines Streptavidin with Horseradish Peroxidase (HRP) and is employed in labeled Streptavidin-Biotin immunoassay systems for antigen detection. This methodology includes sequential incubations of the sample with antigen-specific unlabeled primary antibody, biotinylated secondary antibody (reacting with the primary antibody), enzyme-labeled Streptavidin, and a chromogenic substrate. In Southern or Northern blot analysis, this conjugate also binds to nucleic acids labeled with biotinylated nucleotide analogues.
