名稱	Acarbose
描述	Acarbose (BAY g 5421) is a hypoglycemic agent that is an inhibitor of orally active alpha-glucosidase (IC₅₀=11 nM). Acarbose enhances the hypoglycemic effect of sulfonylureas or insulin.
細(xì)胞實(shí)驗(yàn)	Cell viability is determined using the MTT assay. Cells are seeded in 24-well culture plates at a density of 2×104 cells/well, incubated for 48 h, treated with acarbose at varying concentrations (0.5, 1.0, 2.0, 3.0, and 5.0 μM) for 24 h; or pre-treated with TNF-α (20 ng/mL) for either 24 h or 48 h to evaluate the dose-dependent effects of acarbose on VSMC growth and viability, cultured with 0.5 mg/mL MTT at 37°C in a humidified atmosphere of 5% CO2 for another 4 h, and solubilized with isopropanol. The viable cell number varies directly with the concentration of formazan product measured spectrophotometrically at 563 nm.
體外活性	方法： TNF-α 預(yù)處理的A7r5 細(xì)胞用Acarbose（1、2 和 3 μM）處理后使用Western Blot檢測靶蛋白表達(dá)水平。結(jié)果： Acarbose劑量依賴性地降低 β-半乳糖苷酶、Ras 表達(dá)并增加 p-AMPK 表達(dá)。[1]
體內(nèi)活性	方法：為研究Acarbose的降血糖作用，將Acarbose（30、60 mg/kg）灌胃給大鼠，每天一次，持續(xù)8周。結(jié)果：Acarbose降低空腹血糖，并調(diào)節(jié) DM 大鼠的葡萄糖耐量而不減輕體重。Acarbose 顯著抑制 DM 大鼠血清 IL6 和 TNF-α。[2] 方法：為研究Acarbose對(duì)動(dòng)脈粥樣硬化的影響，將Acarbose（2.5-5.0 mg/kg）喂食給兔子，持續(xù)25周。結(jié)果：Acarbose 顯著且劑量依賴性地降低新內(nèi)膜 IL-6 的強(qiáng)度，TNF-α 和 iNOS 染色，并顯著增加新內(nèi)膜 p-AMPK 染色的強(qiáng)度。Acarbose 顯著且劑量依賴性地降低 HCD 喂養(yǎng)兔的新內(nèi)膜 Ras 和 β-半乳糖苷酶表達(dá)，而體重沒有減輕。[1]
