Bioactivity

名稱	Mertansine
描述	Mertansine is a tubulin inhibitor and an antibody-conjugable maytansinoid alkaloid. The IC₅₀ of Mertansine against HCT-15 and A431 cells is 0.750 and 0.04nM.
體外活性	方法：將HCT-15和A431細(xì)胞與Mertansine (0-3 nM)孵育72小時(shí)，然后將培養(yǎng)基從板中抽離并替換為新鮮培養(yǎng)基。讓培養(yǎng)物在電鍍后生長(zhǎng)并形成菌落共7-10天。將培養(yǎng)物固定，用0.2%結(jié)晶紫在10%福爾馬林/ PBS中染色，計(jì)數(shù)菌落。結(jié)果：Mertansine可以抑制HCT-15和A431細(xì)胞的生長(zhǎng)。[1] 方法：將4T1細(xì)胞與Mertansine（5-100μg/mL)孵育4小時(shí)，CCk8檢測(cè)細(xì)胞活力。結(jié)果：Mertansine可以抑制4T1細(xì)胞的生長(zhǎng)。方法：將MDA-MB-231 細(xì)胞與Mertansine（0.001355–13.55μM）孵育48小時(shí)，MTT檢測(cè)癌細(xì)胞活力。結(jié)果：Mertansine能夠較好的抑制MDA-MB-231 細(xì)胞的增值，IC50值為0.12μM。[3]
體內(nèi)活性	方法：將Mertansine（0.5mg/kg)對(duì)原位4T1腫瘤小鼠的抗腫瘤活性進(jìn)行評(píng)價(jià) 結(jié)果：顯著提高了癌細(xì)胞凋亡，且作用呈劑量依賴性。[2] 方法：使用 MDA-MB-231 三陰性乳腺腫瘤小鼠評(píng)估Mertansine的體內(nèi)治療性能。每3天給予Mertansine，總共注射四次，劑量為0.8mg/kg。結(jié)果：Mertansine表現(xiàn)出適度抑制 MDA-MB-231 三陰性乳腺腫瘤小鼠中癌細(xì)胞的生長(zhǎng).[3]
