名稱	ATI-2341
描述	ATI-2341, pepducin targeting the C-X-C chemokine receptor type 4 (CXCR4), is an allosteric agonist activating the inhibitory heterotrimeric G protein (Gi) to promote inhibition of cAMP production and induce calcium mobilization.
細(xì)胞實(shí)驗(yàn)	Receptors(CXCR4-eGFP) are transiently transfected into HEK-293 cells and are plated 24 h after transfection on glass coverslips coated with poly-D-lysine. The next day, cells are treated with either vehicle alone or with varying concentrations of ATI-2341 for 30 min at 37 °C followed by fixation with methanol for 5 min at −20 °C. GFP fluorescence is visualized directly using a Zeiss Axiovert inverted microscope. Images are processed using Adobe Photoshop and Adobe Illustrator. (Only for Reference)
體外活性	ATI-2341能夠在表達(dá)CXCR4的細(xì)胞中引發(fā)與CXCR4及G蛋白依賴的信號傳導(dǎo)、受體內(nèi)化和趨化性。它是親和力較高的激動劑，具有194 ± 16 nM的EC50值和81 ± 4%的內(nèi)在活性。ATI-2341是一種高效的骨髓PMNs（多形核白細(xì)胞）和HSPCs（造血干細(xì)胞和前體細(xì)胞）動員劑，為HSPCs招募至ABMT（自體骨髓移植）前的治療方法提供了新的策略。同時，ATI-2341能引起CCRF-CEM細(xì)胞的趨化性，產(chǎn)生典型的趨化劑所觀察到的鐘形曲線[1]。
體內(nèi)活性	腹腔內(nèi)給予BALB/c小鼠ATI-2341（i.p.注射）會引起劑量依賴性的PMNs向腹膜的招募，最大效應(yīng)出現(xiàn)在405 nmol/kg時。ATI-2341的更高濃度會導(dǎo)致招募減少，這種現(xiàn)象與化學(xué)趨化劑通常見到的鐘形曲線相似。ATI-2341通過靜脈（i.v.）給藥后，小鼠外周血液中PMNs的數(shù)量呈劑量依賴性增加，在給藥后90分鐘測量達(dá)到最大效應(yīng)，此時ATI-2341的濃度為0.66 μMol/kg。在所有測試劑量下，ATI-2341對淋巴細(xì)胞的動員均無影響[1]。
