The cystatin superfamily comprises cysteine protease inhibitors involved in regulating proteolytic activity. Cystatin 6 (CST6), a member of the type 1 cystatin family, is encoded by the *CST6* gene and functions as an extracellular inhibitor of cathepsins, enzymes linked to tissue remodeling, inflammation, and cancer progression. Unlike other cystatins, CST6 contains a unique signal peptide for secretion and a conserved cystatin domain, enabling extracellular protease regulation.

Research highlights CST6's dual role in cancer biology. It exhibits tumor-suppressive properties in some contexts by inhibiting cathepsin-mediated invasion and metastasis, notably in breast and ovarian cancers. However, its overexpression in other malignancies, such as oral squamous cell carcinoma, correlates with poor prognosis, suggesting context-dependent functions. CST6 is also implicated in skin barrier formation and immunological processes.

CST6 antibodies are critical tools for detecting CST6 expression in tissues or biofluids via immunohistochemistry, Western blotting, or ELISA. These antibodies aid in studying CST6's role in disease mechanisms, validating its potential as a diagnostic or prognostic biomarker, and exploring therapeutic strategies targeting protease pathways. Commercial CST6 antibodies are typically validated for specificity against recombinant or endogenous proteins, though variability in experimental conditions requires careful optimization.
