VEGF165是VEGF-A最豐富的剪接變體，由內(nèi)皮細胞、巨噬細胞和T細胞等多種細胞產(chǎn)生。該因子參與血管生成、血管內(nèi)皮細胞存活、生長、遷移及血管通透性調(diào)節(jié)。VEGF基因表達受缺氧、炎癥細胞因子和癌基因誘導。VEGF165通過與硫酸乙酰肝素結(jié)合，錨定于細胞表面及細胞外基質(zhì)中，并與受體酪氨酸激酶VEGFR1/VEGFR2結(jié)合。作為唯一能結(jié)合共受體神經(jīng)纖毛蛋白-1（NRP-1）和神經(jīng)纖毛蛋白-2（NRP-2）的剪接變體，VEGF165可增強VEGFR2信號傳導。其與VEGFR1/VEGFR2的結(jié)合會誘導PI3K/AKT、p38 MAPK、FAK及樁蛋白信號通路的激活，在多種癌癥的腫瘤血管生成中發(fā)揮關(guān)鍵作用。
