背景介紹

ErbB3（亦稱Her3，即人表皮生長因子受體3）是表皮生長因子受體(EGFR)家族中的受體酪氨酸激酶成員。這種膜結(jié)合糖蛋白雖具有神經(jīng)調(diào)節(jié)蛋白結(jié)合結(jié)構(gòu)域，但其激酶結(jié)構(gòu)域不具備活性功能。因此它能結(jié)合配體，卻無法通過蛋白質(zhì)磷酸化介導細胞內(nèi)信號轉(zhuǎn)導。然而，ErbB3可與ErbB2或其他EGFR家族成員形成異源二聚體，由后者完成酪氨酸磷酸化過程，從而構(gòu)建有功能的受體復合體并激活相關(guān)通路，最終引發(fā)細胞增殖或分化。該蛋白在前列腺癌、膀胱癌及乳腺癌等多種惡性腫瘤中均存在過表達現(xiàn)象。通過選擇性剪接可產(chǎn)生不同亞型，其中缺乏跨膜區(qū)的分泌型異構(gòu)體能調(diào)節(jié)膜結(jié)合型受體的活性。
