人表皮生長因子受體2（HER2）亦稱ERBB2、HER-2、HER-2/neu、NEU、NGL、TKR1及c-erb B2，是一種可導致乳腺癌侵襲性增強的蛋白質(zhì)。該分子屬于ErbB蛋白家族（更常被稱為表皮生長因子受體家族），是一種細胞膜表面結(jié)合的受體酪氨酸激酶，通常參與調(diào)控細胞生長與分化的信號轉(zhuǎn)導通路。HER2被認為是一種孤兒受體，表皮生長因子家族中任何配體均無法將其激活。約30%的乳腺癌存在HER2基因擴增或其蛋白產(chǎn)物過表達現(xiàn)象，該受體在乳腺癌中的過表達與疾病復發(fā)率升高及預后不良密切相關(guān)。研究表明，HER2在發(fā)育過程、腫瘤發(fā)生、神經(jīng)肌肉接頭信號傳遞以及細胞生長與分化調(diào)控中均發(fā)揮重要作用。
