背景介紹
程序性死亡蛋白1（PD-1）亦稱CD279和PDCD1，是一種I型膜蛋白，屬于延伸的CD28/CTLA-4 T細胞調(diào)節(jié)家族成員。PDCD1表達于活化的T細胞、B細胞、巨噬細胞、髓系細胞及部分胸腺細胞表面。PD-1擁有兩個B7家族配體PD-L1與PD-L2。經(jīng)IFN-γ處理后，PD-L1幾乎在所有小鼠腫瘤細胞系（包括PA1骨髓瘤、P815肥大細胞瘤及B16黑色素瘤）中均有表達；而PD-L2表達范圍更為局限，主要存在于樹突狀細胞及少數(shù)腫瘤細胞系。PD-1通過抑制PI3K/AKT通路的激活與轉(zhuǎn)導(dǎo)，進而抑制T細胞增殖及IL-1、IL-4、IL-10和IFN-γ等相關(guān)細胞因子的產(chǎn)生。此外，PD-1共交聯(lián)可通過去磷酸化關(guān)鍵信號轉(zhuǎn)導(dǎo)分子來抑制BCR介導(dǎo)的信號。體外實驗中，用PD-L1-Ig處理抗CD3刺激的T細胞可導(dǎo)致T細胞增殖與IFN-γ分泌減少。目前正在研發(fā)靶向PD-1的單克隆抗體，通過增強免疫系統(tǒng)功能用于癌癥治療。
