Englumafusp alfa is an investigational bispecific antibody-like fusion protein developed by Roche. It is engineered as a T-cell engager with integrated co-stimulation. Its unique design allows it to simultaneously bind to CD19 on B cells (common in B-cell malignancies) and 4-1BB (CD137) on T cells. By bridging these two cell types, it achieves two key functions: first, it redirects and activates T cells directly against CD19-positive tumor cells; second, and crucially, its engagement with 4-1BB delivers a potent co-stimulatory signal to the same T cell. This dual action is designed to enhance the tumor-killing activity, proliferation, and survival of the engaged T cells, potentially creating a more robust and durable anti-tumor immune response compared to T-cell engagers without co-stimulation.
