Etigilimab (development code MPH-313) is an investigational, fully human monoclonal IgG1 antibody developed for cancer immunotherapy. It functions as an antagonist of the T-cell immunoglobulin and ITIM domain (TIGIT) receptor, an emerging immune checkpoint. By binding to TIGIT with high affinity, etigilimab blocks its interaction with its primary ligands, CD155 (PVR) and CD112 (PVRL2, nectin-2), which are commonly upregulated on tumor cells and antigen-presenting cells within the tumor microenvironment. This blockade prevents TIGIT from transmitting an inhibitory signal to T cells and natural killer (NK) cells, thereby "releasing the brake" on these immune effector cells. This aims to enhance their anti-tumor activity, including proliferation, cytokine production, and cytotoxic function.
