亨德拉病毒(HeV)和尼帕病毒(NiV)是于20世紀(jì)90年代中后期發(fā)現(xiàn)的亨尼帕病毒,具有廣泛的宿主嗜性,可導(dǎo)致人類和動(dòng)物出現(xiàn)嚴(yán)重且通常致命的疾病。HeV 和 NiV 通過兩種包膜糖蛋白的協(xié)同作用感染宿主細(xì)胞:G 糖蛋白附著于細(xì)胞受體,從而觸發(fā)融合(F)糖蛋白執(zhí)行膜融合功能。G 蛋白是 II 型同源四聚體跨膜蛋白,負(fù)責(zé)與 ephrinB2 或 ephrinB3(ephrinB2/B3)受體結(jié)合。F 蛋白是 I 型同源三聚體跨膜蛋白,以未成熟的前體 F0 形式合成,并在內(nèi)吞再循環(huán)過程中被組織蛋白酶 L 切割,產(chǎn)生成熟且由二硫鍵連接的 F1 和 F2 亞基。當(dāng) NiV G 蛋白與 ephrinB2/B3 結(jié)合后,其構(gòu)象發(fā)生變化,從而觸發(fā) F 蛋白,并將 F 蛋白的疏水性融合肽插入靶膜。隨后,F 蛋白重新折疊為更穩(wěn)定的融合后構(gòu)象,驅(qū)動(dòng)病毒與宿主細(xì)胞膜的融合,形成一個(gè)孔道,以便將病毒基因組遞送入細(xì)胞質(zhì)。
