Synthesis of Simple N-hydroxymethyl Isatin
The N-hydroxymethyl isatin was synthesized as shown inScheme (2). 1 mmol isatin was dissolved in 20ml methanolin a flask. Then 1.2 mmol formaldehyde was added dropwise.The mixture was stirred under refluxing until the disappearanceof isatin, as evidenced by thin-layer chromatography.The solvent was removed in vacuo and the residuewas separated by column chromatography (silica gel, petroleumether/ethyl acetate = 4: 1), giving N-hydroxymethylisatin (compound 13).
