(6) 4.5g (14.59mmol) of methyl 2-amino-5-methoxy-4-[3-(pyrrolidin-1-yl)propoxy]benzoate were added to a 100mL there-necked flask, 5mL of glacial acetic acid and 10mL of water were added with 1.4g (21.88mmol) of sodium cyanate at room temperature, and reacted at 65°C for 10h, cool to room temperature, add 10 mL of methanol and 20 mL of water, adjust the pH value to 9 with sodium hydroxide solid, and continue the reaction at 65 °C for 2 h. The methanol was distilled off under reduced pressure, filtered with suction, and the obtained filter cake was washed twice with 10 mL of water and dried. Obtained 6-methoxy-7-[3-(1-pyrrolidinyl)propoxy]-2,4(1H,3H)-quinazolinedione (white solid, 4.13 g, yield 88.6%).
