Steps:

Firstly, Histidine (9.30 g,60.0 mmol) and methanol (100 ml) were mixed in the 250 ml roundbottom flask. Under the conditions of the magnetic stirring and ice bath,SOCl2 (10.0 g, 85 mmol) was dropwise added into the mixtures. Theresulting reactants were kept at reflux temperature overnight. Then, thesolvent was evaporated by vacuum rotary evaporation. The rude productwas dispersed into the solvent dichloromethane and SOCl2 was removedby evaporation. The white product of Histidine methyl ester hydrochloride (12.1 g) was obtained with the yield of 83%.
