Synthesis of 5-(4-methylpiperazin-1-yl)-1,3,4-thiadiazol-2-amine (Precursor-21)
5-Bromo-1,3,4-thiadiazol-2-amine 40 (2.20 g, 12.29 mmol) and 1- methylpiperazine (2.46 g, 24.6 mmol) were heated in n-propanol (30 mL) in a sealed tube at 100°C for 8 h. The reaction was cooled to rt, solvents evaporated under reduced pressure and the crude obtained was triturated with MeOH to obtain 5-(4- methylpiperazin-l-yl)-1,3,4-thiadiazol-2-amine precursor-21 (1.3 g, 53% yield) as an off white solid. MS: 200.16 (M+H).
