Berberine hydrochloride (1, 20g, 62.11mmol) was placed in the round bottom flask and in a vacuum, stirred at 180°C for 20min to give the crude product, which was purified using flash silica gel column chromatography (CH2Cl2/MeOH=10:1) to give 22g of the red solid compound berberrubine (2, 98% yield). 1H NMR (400MHz, DMSO-d6): δ 9.22 (s, 1H), 8.06 (s, 1H), 7.56 (d, 1H, J=8.0Hz), 7.43 (s, 1H), 6.96 (s, 1H), 6.85 (d, 1H, J=8.0Hz), 6.10 (s, 2H), 4.70 (t, 2H, J=6.0Hz), 3.78 (s, 3H), 3.23 (t, 2H, J=6.0Hz). MS (ESI+) m/z: 322 [M+H]+.
