Steps:
1-(Carboxymethyl)cyclopentanecarboxylic acid (4b).
A solution of potassium cyanide (0.8 g, 12.3 mmol) in water (2 ml) was added to a solution of ethyl 2-cyano-2-cyclopentylideneacetate (2b) (2.0 g, 11.5 mmol) in ethanol (40 ml). The reaction mixture was heated at reflux for 5 hours. After cooling to room temperature, the reaction mix-ture was concentrated to dryness. The residue was cautiously treated with cc. HCl (15 ml), and this acidic water phase was extracted with ethyl acetate (3×30 ml). The organic extract was dried (MgSO4), filtered, and concentrated. The residue was dissolved in cc. HCl (12 ml) and heated at reflux for 16 hours. After cooling to room temperature the precipitated crystals were filtered, and dried to give 0.7 g white solid. Y = 36%, IR: 1702 cm -1 (s), 1 H NMR (799.7 MHz, DMSO-d 6 ) δ = 1.48-1.62 (m, 6H), 1.98-2.03 (m, 2H): 4× cyclopentane CH 2 , 2.54 (s, 2H, H 2 -1’), 12.0 (br, COOH) ppm; 13 C NMR (201.1 MHz, DMSO-d 6 ) δ = 24.7 (C-3, C-4), 36.0 (C-2, C-5), 42.4 (br, C-1’), 49.8 (C-1), 172.8 (C-2’), 177.9 (C-3’) ppm; HRMS: M+H=173.08076 (delta=-0.4 ppm; C 8 H 13 N 4 ). HR-ESI-MS-MS (CID=35%; rel. int. %): 155(82); 154(100); 141(5); 127(46); 110(9); 100(14); 84(6).
