Steps:
Synthesis of 1- (4-morpholinophenyl)guanidine (7)
Concentrated hydrochloric acid (4 mL) was added to a solution of 4-morpholinoaniline (1.78 g, 10 mmol) in EtOH (10 mL) at 0 °C. Then cyanamide (2 mL of 50% solution in water, 25.7 mmol) was added and the mixture was heated to reflux for 10 h. After cooling to room temperature, the solvent was removed under reduced pressure. Then saturated sodium carbonate aqueous solution (10 mL) was added to the residue. The solid was filtered off, washed with acetone and dried under reduced pressure to afford compound 7 as: Off white solid; yield 2.3 g (82%);16 m.p. 224-226 °C; IR: 3084, 1693, 1631, 1518, 1450, 1406, 1267 cm-1. Anal. calcd for C11H16N4O: C, 59.98; H, 7.32; N, 25.44; found: C, 59.87; H, 7.49; N, 25.59%. ESI-MS: 221.2 [M + H]+, 219.1 [M - H]-; 1H NMR (300 MHz, DMSO-d6): δ 7.16 (brs, 2H), 6.86-6.89 (m, 4H), 6.59 (brs, 1H), 4.25 (brs, 1H), 3.71-3.74 (m, 4H), 3.02-3.05 (m, 4H); 13C NMR (75 MHz, DMSO-d6): δ 160.0, 154.9, 147.7, 124.7, 116.1, 66.1, 48.9.
