3,5,5,8,8-五甲基-5,6,7,8-四氫萘-2-胺(6)的合成步驟如下:將1,1,4,4,6-五甲基-7-硝基-1,2,3,4-四氫化萘(5)(2.5g,10.1mmol)溶解于乙酸乙酯(205mL)中,配制成0.05M的溶液。使用ThalesNano H-cube?反應(yīng)器,在70°C和2-5巴壓力下,以1.0毫升/分鐘的流速,通過10毫升Pd/C濾芯進(jìn)行氫化反應(yīng),重復(fù)兩次。反應(yīng)完成后,將所得溶液在真空下濃縮,得到黃色結(jié)晶固體的目標(biāo)產(chǎn)物6(2.13g,收率97%),熔點(diǎn)為205°C。產(chǎn)物的結(jié)構(gòu)通過以下表征數(shù)據(jù)確認(rèn):1H NMR(400MHz,CDCl3)δ 7.00(s,1H),6.63(s,1H),3.34(br s,2H),2.15(s,3H),1.63(s,4H),1.26(s,12H);13C NMR(100.6MHz,CDCl3)δ 143.5,142.0,135.2,128.3,120.5,112.6,35.3,35.2,33.8,33.4,32.0,31.8,17.1;IR(neat)ν 3404,3335,2956,2925,1626,1504 cm-1;LC-MS-CI(M+H)+計(jì)算值C15H24N 218.1909,實(shí)測(cè)值218.1908。
