以2-氨基-4-氟苯酚(10g,78.8mmol)、二硫化碳(35mL)和氫氧化鉀(5.3g,94.6mmol)為原料,在乙醇(200mL)中混合,加熱至回流反應(yīng)過夜。反應(yīng)完成后,將反應(yīng)混合物濃縮至干,隨后用水稀釋。用1M鹽酸溶液中和稀釋后的溶液,再用乙酸乙酯進(jìn)行多次萃取。合并有機(jī)相,用飽和食鹽水洗滌,無水硫酸鈉干燥,最后在真空條件下濃縮,得到5-氟苯并[d]惡唑-2-硫醇9.3g,收率70%。產(chǎn)物經(jīng)1H NMR(300MHz,DMSO-d6)鑒定:δ7.51(ddd,J=8.8,4.2,0.6Hz,1H),7.15-7.04(m,2H),3.31(brs,1H)。LC-MS(m/z):170.2。
