一般步驟:以2-甲基噻吩和乙酰氯為原料,采用Friedel-Crafts?;磻?yīng)合成2-乙酰-5-甲基噻吩。具體操作如下:在裝有磁力攪拌子的雙頸圓底燒瓶中加入預(yù)先制備的MoO4(AlCl22(5wt%,10mg))催化劑,該催化劑需在150℃下活化2小時。隨后,向反應(yīng)瓶中加入2-甲基噻吩(1g,9.26mmol)和乙酰氯(5當(dāng)量)。反應(yīng)混合物在室溫下攪拌,直至反應(yīng)完成,反應(yīng)進(jìn)程通過薄層色譜法(TLC)監(jiān)測。反應(yīng)完成后,過濾反應(yīng)混合物以分離固體催化劑,該催化劑可回收用于下一次Friedel-Crafts?;磻?yīng)。反應(yīng)混合物依次用二氯甲烷(DCM)和水洗滌,此過程重復(fù)三次。收集有機(jī)層,用無水硫酸鈉干燥,隨后在旋轉(zhuǎn)蒸發(fā)器上濃縮,得到粗產(chǎn)物。最后,通過柱色譜法對粗產(chǎn)物進(jìn)行純化,得到目標(biāo)化合物2-乙酰-5-甲基噻吩。
