實施例1a 6-甲基-2-(4-甲基苯基)咪唑并[1,2-a]吡啶-3-乙酸的合成:在0℃及氬氣保護下,將唑吡坦(8.0 g,26.0 mmol)溶于6N鹽酸(80 mL)中,磁力攪拌。將反應(yīng)混合物加熱回流18小時。反應(yīng)完成后,將混合物冷卻至0℃,緩慢加入20%氫氧化鈉溶液調(diào)節(jié)pH至6.5。析出的白色固體經(jīng)真空過濾收集,得到目標(biāo)產(chǎn)物6-甲基-2-(4-甲基苯基)咪唑并[1,2-a]吡啶-3-乙酸(6.9 g,收率94%),為白色固體。1H NMR (DMSO-d6) δ 2.30 (s, 3H), 2.35 (s, 3H), 3.94 (s, 2H), 7.10 (d, J = 9.1 Hz, 1H), 7.25 (d, J = 7.8 Hz, 2H), 7.48 (d, J = 9.1 Hz, 1H), 7.72 (d, J = 7.8 Hz, 2H), 8.18 (s, 1H)。13C NMR (100 MHz, DMSO-d6) δ 18.4, 21.4, 30.8, 114.8, 116.6, 121.6, 122.9, 127.9, 128.2, 129.8, 132.4, 137.3, 143.0, 143.5, 171.8。
