以2-(乙酰乙酰氨基)苯甲酸甲酯為原料合成3-乙?;?4-羥基喹啉-2(1H)-酮的一般步驟:向100 mL圓底燒瓶中加入0.43 g(18.8 mmol)金屬鈉和30 mL無水甲醇,攪拌至金屬鈉完全溶解。隨后加入20 mL無水二氯甲烷,攪拌均勻后,加入4.43 g(18.8 mmol)2-(乙酰乙酰氨基)苯甲酸甲酯,確保混合均勻。將反應(yīng)混合物加熱回流5小時,然后冷卻至室溫。向反應(yīng)混合物中加入50 mL冰水,并用稀鹽酸調(diào)節(jié)pH至強酸性,此時有大量白色固體析出。通過過濾收集白色固體,依次用水和乙醇洗滌,干燥后得到3.66 g白色固體產(chǎn)物3-乙?;?4-羥基喹啉-2(1H)-酮,熔點為256-257℃,收率為95.7%。
