通用方法:在0℃下,將叔丁醇鉀(t-BuOK,2.47g,22.00mmol)分四批加入到甲基三苯基溴化鏻(7.12g,20.00mmol)的無水四氫呋喃(THF,70mL)溶液中。將反應(yīng)混合物在0℃下攪拌2小時(shí),隨后在30分鐘內(nèi)緩慢滴加4-氟苯乙酮(20mmol)的無水THF(20mL)溶液。滴加完畢后,將反應(yīng)混合物在室溫下繼續(xù)攪拌16小時(shí)。反應(yīng)完成后,用飽和氯化銨(NH4Cl,50mL)溶液淬滅反應(yīng)。將反應(yīng)液用石油醚(3×50mL)萃取三次。合并所有有機(jī)相,用飽和食鹽水(50mL)洗滌,無水硫酸鈉干燥,過濾并濃縮。最后,通過快速柱色譜法純化殘余物,以石油醚(Pet)作為洗脫劑,得到目標(biāo)產(chǎn)物1-氟-4-(丙-1-烯-2-基)苯。
