以2-羥基-3-硝基苯甲酸甲酯為原料合成3-氨基-2-羥基苯甲酸甲酯的一般步驟如下:在室溫條件下,將2-羥基-3-硝基苯甲酸甲酯(18.1g,93mmol)與5%鈀碳(5g)在甲醇(200mL)中的懸浮液置于帕爾瓶中,在氫氣(60psi)氛圍下?lián)u動(dòng)49小時(shí)。反應(yīng)完成后,通過硅藻土塞過濾反應(yīng)混合物,濾液經(jīng)減壓濃縮除去溶劑,得到黃色固體狀的3-氨基-2-羥基苯甲酸甲酯(15.6g,產(chǎn)率> 99%)。產(chǎn)物結(jié)構(gòu)經(jīng)1H NMR(CDCl3)確認(rèn):δ 10.87(s,1H),7.23(dd,J = 7.7Hz,2H),6.87(t,J = 9.9Hz,1H),3.95(s,3H),3.80(br s,2H);ESI MS m/z 168 [C8H9NO3 + H]+。
