以5-氯-1H-苯并[d][1,3]噁嗪-2,4-二酮和碘甲烷為原料合成5-氯-1-甲基-1H-苯并[d][1,3]惡嗪-2,4-二酮的一般步驟:將碘甲烷(2 mL,4.6 mmol,4當(dāng)量)和N,N-二異丙基乙胺(DIPEA,1 mL,0.74 mol)在室溫下懸浮于N,N-二甲基乙酰胺(DMA,10 mL)中,攪拌10分鐘。隨后加入5-氯靛紅酸酐(200 mg,1.01 mmol,1當(dāng)量),將反應(yīng)混合物在40℃下攪拌5小時。反應(yīng)完成后,過濾收集所得產(chǎn)物5-氯-1-甲基-1H-苯并[d][1,3]惡嗪-2,4-二酮,并用蒸餾水洗滌,得到純品(150 mg,收率70%)。產(chǎn)物的熔點(diǎn)為224℃。紅外光譜(KBr)顯示特征吸收峰υ1774, 1716, 1593 cm^-1。1H NMR(DMSO-d6, 300 MHz)δ7.78(t,J = 8.1 Hz,1H),7.42(m,2H),3.45(s,3H)。元素分析結(jié)果:C9H6ClNO3的計(jì)算值:C,51.08;H,2.86;Cl,16.75;N,6.62;O,22.68。實(shí)測值:C,51.12;H,2.36;Cl,16.71;N,6.58。
