以2,6-二氯-3-羥甲基吡啶為原料合成2,6-二氯-3-吡啶甲醛的一般步驟如下:在26℃下,向(2,6-二氯吡啶-3-基)甲醇(1.0g,5.62mmol)的二氯甲烷(10ml)溶液中加入Dess-Martin試劑(4.8g,11.24mmol)。加完后,將反應(yīng)混合物在室溫下攪拌2小時(shí)。反應(yīng)完成后,通過(guò)加入5% Na2S2O3水溶液淬滅反應(yīng),并繼續(xù)攪拌30分鐘。用二氯甲烷(2×30ml)萃取反應(yīng)混合物。合并有機(jī)層,依次用飽和Na2S2O3溶液(50ml)、鹽水(30ml)洗滌,無(wú)水Na2SO4干燥,減壓濃縮,得到粗品2,6-二氯-3-吡啶甲醛(800mg,收率80%),直接用于下一步反應(yīng)。產(chǎn)物經(jīng)1H NMR(400MHz,CDCl3)表征:δ 10.38(s, 1H),8.19(d, J = 8.0Hz, 1H),7.44(d, J = 8.0Hz, 1H)。
