以2-氨基苯并噻唑為原料合成2-肼基苯并噻唑的一般步驟:在5℃條件下,將水合肼(1當(dāng)量)溶解于乙二醇中。在攪拌下,緩慢滴加濃鹽酸(2mL),隨后加入2-氨基苯并噻唑(1當(dāng)量)。將反應(yīng)混合物加熱回流2-3小時,之后冷卻至室溫。反應(yīng)完成后,過濾收集形成的固體產(chǎn)物,用水洗滌,并通過乙醇重結(jié)晶純化,得到目標(biāo)化合物2-肼基苯并[d]噻唑(32)。產(chǎn)物為白色晶體,收率90%,熔點范圍為200℃至202℃。1H NMR(DMSO-d6, 400MHz)數(shù)據(jù)如下:δ 4.84(2H, s),6.85-7.65(4H, m),8.62(1H, s)。ESI-MS(m/z)分析顯示分子離子峰為166(MH+)。
